Its not a nipple like you see on ball-joint or other front end parts IIRC it's called a fish eye, you need a grease needle to get grease in the opening, its not easy to find or get grease in it, that is why on my 2006 2500 I finally used orange paint to mark its location, I've had a few dealer lube techs tell me it didn't to have one, when it happened I spoke up to service m manager who got there Diesel tech to show them how to find it...

Also make sure it is clean and free of dirt before you attempt to put grease in.

Good move on the paint. They are tough to find.

I worked as a tech at a Chrysler dealership and we lubed them all the time during oil changes, they are definitely there on a 2500/3500, comes stock from the factory like that. If it doesn't have one it's probably been replaced with an aftermarket shaft or joint. like others said it's a little tricky to find but it faces forward and once you know what you're looking for it'll be easy to find in the future. You absolutely need to lubricate those though, in the 4 years I worked there I saw at least 4 or 5 trucks that had grenaded transfer cases because of failed drive shaft joints from lack of lubrication (ask me for pictures, they are crazy). A quick google/youtube search will help with that, and you'll need a grease gun needle attachment to lubricate it as well, not the ones that have needle points you can poke into boots, but just a round tip. It looks like this.
