Diagnosing cam and lifter

Hey fellas I want to watch my cylinder 3 rocker arm and valve spring to figure out if my cam and lifter are toast. A few questions if I pull the intake manifold to make it easier to access the valve cover and pull it as well. Can I just pull the fuel relay fuse and turn the truck over to watch the motions. What's the best procedure should I remove the spark plugs as well? Can I turn it over with the intake manifold off as well? Any tips greatly appreciated. 2011 ram 1500 sport 5.7 and yes I have an external fuel pump relay fuse as of course my TIPM is malfunctioning... (on my second one with a failed fuel pump relay haha)

Doesn't hurt to pull the plugs.A better option is to pull the starter relay and jump it,that way nothing gets power like the coils,fuel pump etc,and you can watch the engine turn over with-out needing somebody to run the ignition.

You do not need to remove the intake manifold to make it easier to remove the valve covers. They come off at an angle free and clear (see pic), though you may have a little less clearance on the driver's side. Just remove the valve cover and do what @Wild one suggested above, this way you can just stare at it til you see what you need to see.

It would appear to be a dead cylinder on the intake side most likely a lifter i just checked the pushrod it's fine.
