I have 2004 5.7 that the knock sensor wires burned up on one side, will a truck start and run with a missing / disconnected knock sensor?

Yes, on this generation of Hemi the knock sensor is for misfire detection. You may get a CEL due to the loop being incomplete. I'd suggest either repairing or replacing the damaged wiring ASAP though.
