My 2000 Ram 1500 recently started making a clicking sound while driving that sounds like its coming from the rear driver side, the rhythm matches with the trucks speed. I jacked up the rear axle and spun both rear wheels with the wheels on and off, no sound. Do any of you guys know what could be causing this?

Are you sure it's not the front clicking? Could be an axle u-joint.

4x4? The front, outer u-joints make a very distinct "clacking" sound when they're toasted, especially at parking lot speeds, and the noise will increase in frequency until you get up to around highway speed, where they'll suddenly go quiet and everything seems okay...until you slow down to just the right speed, and you'll start hearing them clacking again.

That would be it, I checked the outer left axle u-joint and its completely fried. Thanks for the advice.
