I have a 2000 Ram 1500 4x4. I noticed going down the highway every time I let off the gas pedal I would feel a vibration for about 1.5 seconds. It felt like it came from the tranny. Could anyone give me some more insight on this issue?

I was told that that could be coming from the overdrive gear. Mine has done that before but I haven't noticed it in awhile now.

I second the u-joint. I had the same symptom when my rear u-joint was starting to go bad. If I gave it a little pedal it would go away.

Carrier bearing? Maybe yer 4th gen needs it! 2nd gens had 'em but a 2000 1500 4wd is usually a 1 piece drive shaft, even an 8' bed is usually 1 piece.
Checking U-joints is sage advice.
Damn Bro i didn't even notice it was a 2000 lol ... sorry
