2011 ram 1500 4x4 5.7 140k miles.

Any thoughts on the above kit. Will the included tuner be all I need to tune and delete mds?

Is the upgrade to Mopar lifters worth the extra $?

Is removing the pan necessary as it's 4x4 and looks like a lot more work. The rest of the engine work is too but manageable.

If there are better kits please post them.

There's better cam kits then that one.Check with Flying Ryan Performance/FRP or MMX or Matt Fikac at Moes.
If you do go for that kit,suck it up and get the OEM lifters.How many miles are on the truck,if it's a higher milege truck,you might want to consider installing new valve springs and valve seals,both do wear out over time.
Get a cheap 13 mm wrench that you can bend/modify to get at the oil pick-up tube. Dental floss wrapped around the bolt so when you pull on it,it turns the bolt righty tighty makes it a little easier to get the bolt back in place,be very careful you don't mess up the o-ring on the pick-up tube,otherwise you might not have much in the way of oil pressure.
A straightened out coat hanger or a piece of welding rod,laid lengthwise on the head under the pushrods,helps hold them in place to re-install the rocker shafts/arms.
Pay attention to the head gaskets,they are side specific

Has 140k.

I'd seriously consider new valve springs at that milege.Valve springs do wear out/fatigue with time and milege

Since heads will be off good idea. Do I need special valve spring compressor. Mine dates back to working on 360 and a 383.
